    Big news! 🥳🥂 In July 2025, I will be joining the faculty of University of Pennsylvania, @Penn_CBIO. The Goodman Lab for Synthetic Immunology will combine synthetic biology, genome engineering, and ML-driven functional genomics to study and manipulate primary immune cells. 🧵1/3

    Just put up a new preprint w/ my @geochurch-lab colleague @Schubes0 & team. We show that ssDNA produced in vivo can be used for efficient barcoded genome editing. It scales to pools of millions of genomic edits! (1\6)

    It is mind-blowing that this works at all. But to be clear, this is not reading the cell's transcriptome, just taking a high dimensional image of the cell itself and mapping that to transcriptome training data. 1/2
    No sequencing? No problem! You can shoot lasers at cells and read their transcriptomes at a single cell resolution (ht @AlexandreTrapp ) biorxiv.org/content/10.110…

    Really cool new paper combining retrons + CRISPR spacer aquisition to record transcriptional events as sequential genomic barcodes. Some very impressive DNA jockeying. Retrons are the future. 😁 Congrats @seth_shipman @Schubes0 @jeffnivala @BhattaraiKline!

    Not really evolution, but here's my fav: the synthetic genome assembly map I made in #Python + @ProcessingOrg for our 2013 GRO paper (science.org/doi/10.1126/sc…). This bad boy took more work than any fig I've made since, and in the end it ended up buried in the supplement! 😂
