Abstract
The COVID-19 pandemic created an urgent need for effective early outpatient treatment strategies that could reduce viral replication before severe illness develops. This whitepaper explains the biological mechanism underlying the Zelenko Protocol, an early treatment strategy centered on increasing intracellular zinc concentrations using zinc ionophores and supportive nutrients. SARS-CoV-2 replication depends on the activity of RNA-dependent RNA polymerase (RdRp), the viral enzyme responsible for copying the virus’s RNA genome. Experimental studies have demonstrated that elevated intracellular zinc concentrations can inhibit the coronavirus RNA-dependent RNA polymerase (RdRp) and interfere with viral RNA synthesis. However, zinc ions cannot efficiently cross cell membranes on their own. Zinc ionophores—including hydroxychloroquine, quercetin, and epigallocatechin gallate (EGCG)—can transport zinc into cells and increase intracellular zinc concentrations. This paper explains how viral entry, replication, and assembly occur in SARS-CoV-2 infection and describes how zinc and ionophores interfere with the replication process. It also summarizes the components of the Zelenko Protocol and their roles in supporting immune function and reducing viral replication.
