Are you talking about custom styles? There should be styles applied to the tooltip. See screenshot 4 for an example at https://wordpress.org/plugins/wp-glossary-hover/screenshots/

The screenshot shows the plugin working with the default settings and a default WordPress theme.
Can you try disabling all of your other plugins to see if the problem persists?
I am not using the default WordPress theme, thus this is likely my problem.

Well, to narrow down the problem, can you do each of the following:
Can you try reinstalling the plugin?
Does the problem still persist with all other plugins disabled?
Does the problem still persist when using a default WordPress theme?
