I try my best to follow canon when it comes to how I play Snape. I won't be perfect but I'm going for who he was in the books (and borrowing scenes from Harry Potter and the Deathly Hallows Part II).
First and foremost - Severus Snape is an asshole. He is so let's just get that out there right now. He was willing, at first, to let Harry and James die in exchange for Lily. That takes some serious self-centeredness - he was willing to let a baby be murdered. Yeah, nice guy, right?

Snape's Worst Memory
I hate how the film dealt with this part - totally gutted it. BUT - what they did do was well done. Alan Rickman brings a nice mix of anger and worry to the scene about what will happen if Harry doesn't learn Occulemency. It's just too bad it was too short - no Lily, WTF?! OotP was one of my favorite books in the series but it was one of my most hated movies of the series.
The Dark Lord Descending
Why the film makers cut out this scene I will NEVER understand. Of course, the reasoning behind most of the decisions they made regarding 'Half Blood Prince' will forever boggle my mind. This is a deleted scene from HBP. It is an absolutely breath-taking blend of cinematography, special effects, music, and acting. Without words Alan Rickman conveys the weight of Snape's burden - knowing what he has to do, wondering if he can possibly live with himself, and fearing he won't have the strength and courage necessary for the path that lies before him.
A path that he will have to walk alone once Dumbledore dies.
ALWAYS - Lily and Snape
Don't watch this unless you have read Deathly Hallows or have seen Deathly Hallows Part Two! This, right here, this is why I play Severus Snape. He was a pathetic, sad, angry, lonely little boy who became something more - something great. He was transformed by love and love is the overarching theme of the Harry Potter series - that love makes anything possible. Love changed the path of Snape's life. It was Lily's ability to see the beauty in others when they themselves couldn't see it that allowed Lord Voldemort to be defeated. Lily's love, kindness, and strength were what Severus Snape and Harry Potter leaned on and they were what allowed both men to become something more than they could have ever become on their own. Without Lily Severus would never have made his promise to Dumbledore and Harry would have been lost along with the entire world. Without love we are all lost.
Harry Potter - Dueling Club
Because I cannot get enough of this scene. This is also an excellent example of Snape's superior dueling abilities. I find it highly amusing how both Harry and Draco imitate Snape's stance as opposed to Lockhart's.
Deleted Harry Potter Potions Scene
Yet another scene that was cut short in the movie but thankfully was left intact for bonus features on the DVD. It's just too awesome for words.
Goblet of Fire deleted scene
As someone in the comments said I love how "Karkaroff is all concerned about the Dark Mark and Snape's just trying to cock-block as many kids as possible". Also, there is something about Snape throwing his left arm up out of Karkaroff's reach that gives me chills. Don't ask cause I don't know why.

I love Alan Rickman and J.K. Rowling's thoughts on Snape. Alan sums it up quite well, "What does this word 'loyalty' mean?" Snape's story has always been about trust and loyalty - Alan gets that and that is why he plays the part so perfectly.
