mouse buttons (+ keyboard modifiers) should be mappable as keyboard shortcuts to commands (e.g. undo/redo, toggle symmetry, flip etc.)
toggle tool with mouse buttons (execute command) or "quick tool" + trigger tool with mouse button (just like the right-click customization, instant eyedropper, pen, eraser, etc. with X1 or X2)

I am willing to implement this if I get a green light.

This set of features is too complex to be implemented at the moment (and mainly for newcomers). Even more we are planning new features for the v1.3 which is being developed privately for the next big release.

Anyway if you want to help the first thing would be to possibility to associate X1 and X2 buttons just like it's already possible with right-click configuration (Edit > Preferences > Right-click). Some years ago I started a branch (now abandoned) to click a tool in the toolbar to associate the tool with a specific mouse button i.e. in the same way left/right mouse buttons change FG/BG colors, this could be the same for the toolbar (e.g. clicking with X1 or right-click or middle-click, will associate that specific mouse button with that tool).
