She is the daughter of actors Sean Penn and Robin Wright, and the niece of musician Michael Penn and actor Chris Penn. Dylan is the granddaughter of actor and director Leo Penn and actress Eileen Ryan (born Eileen Annucci). Her brother is actor Hopper Penn. She is a cousin of actor and model Charlie Wright.

Dylan’s paternal grandfather was Leo Z. Penn (the son of Moische/Maurice Daniel Penn and Elizabeth Marion Melincoff). Leo was born in Lawrence, Essex, Massachusetts, to Lithuanian Jewish/Russian Jewish parents.
