    Excited to share a new major capability from Diffuse Bio: RamaX Opt, our ultra-fast affinity maturation and sequence optimization workflow. We demonstrate 4–50× affinity improvements across 4 starting CD3 binders, plus sequence liability removal — all in ~1 week. To our knowledge, this is the fastest affinity maturation method available, outperforming even the most advanced existing experimental and AI approaches. And this is just a limited first pass — single point mutations for some sequences and modest combinatorial diversity for others. RamaX can handle very large and diverse libraries, so there's plenty of room to push further. With DiffuseSandbox for design, RamaX for discovery, and now RamaX Opt for optimization, the full path from target to optimized lead candidate takes weeks, not months. The bigger picture for us at Diffuse Bio: the bottleneck in AI protein design isn't model architecture — it's data. Every affinity maturation campaign on RamaX produces thousands of sequence-affinity measurements that flow directly back into training our next generation of protein design models.     Announcing a unified pipeline for end-to-end protein binder design and experimental validation at scale 🤖🤝🏾🧬 DiffuseSandbox ➡️ RamaX Computational protein design hits a bottleneck at the lab bench: there is friction between design generation and validation. Even with SOTA models, we’ve seen that difficult therapeutic targets often require screening thousands of candidates to find a functional hit. In DiffuseSandbox, you can now generate up to 10,000 designs and then with one click immediately request experimental screening with RamaX, with data back in 1-2 weeks!     We're excited to unveil RamaX: our platform for ultra-fast, high-sensitivity screening of massive binder libraries (100K to 1B minibinders, VHHs, or scFvs) in just 1-2 weeks. RamaX enables push-button protein design: start with a naive or AI-designed binder library and receive validated binders in weeks.  RamaX matches yeast surface display in sensitivity and selectivity while being up to 8x faster and enabling multiplex screening against many antigens simultaneously.
