Researchers Engineer CAR T Cells In Vivo against Multiple Malignancies | Cancer Discovery | American Association for Cancer Research
Abstract:. Researchers utilized a two-vector system, consisting of an enveloped delivery vehicle and an adeno-associated virus to deliver CRISPR tools and a large DNA payload, to achieve stable and cell-specific expression of chimeric antigen receptor (CAR) T cells. This study demonstrates the first in vivo engineering of CAR T cells at a therapeutic level for both hematologic and solid cancers, and opens the door for more efficient, precise, and affordable CAR T-cell therapies.

A massive study spanning 185 countries concludes that about four in 10 cancer cases are linked to modifiable risk factors such as smoking, infections, and alcohol consumption. Stratifying by sex, researchers found three in 10 cancer cases in women and one in two in men were due to modifiable risk factors. The findings “reinforce the call for effective cancer prevention,” the researchers say.

Base Editing Might Be Key to CAR T-cell Therapy
Researchers used a unique base-editing strategy to engineer chimeric-antigen receptor (CAR) T cells that engraft in the host and remain invisible to remaining antibody drugs. Of the 11 patients with T-cell acute lymphoblastic leukemia who received these BE-CAR7 T cells, all had complete morphogenic remission after 28 days, 82% underwent stem-cell transplantation, and 64% remain in remission 3 to 36 months after treatment.
